Cytochemical evaluation was conducted on sperm succinate dehydrogenase
(SD) in crossbred, Holstein and Murrah buffalo. The enzymic action on
disodium succinate hexahydrate (mol. wt 270) and disodium succinate (
mol. wt 162) were tried at 0.2M and 0.5M concentrations. The disodium
succinate (anhydrous) yielded better results for the cytochemical appr
aisal of SD. Also, the other sperm reductases confounding the assessme
nt of SD in situ, were better blocked by desiccation (for nearly 10 mi
n at room temperature) than the heat treatment (at 50-degrees-C for 30
min).
