Synthesis of a beta-D-glucan decasaccharide, with alternating beta-1,3
and beta-1,4-linkages, is described. The decasaccharide is fully C-de
uterated except for the central cellobiose residue. The main glycosyla
tion methods used were activation of glycosyl bromides with silver tri
flate or activation of thioglycosides with dimethyl(methylthio)sulfoni
um triflate (DMTST). In order to establish the anomeric configuration
of the various oligosaccharide derivatives, a fully C-protonated decas
accharide was also synthesized by a parallel route.
