T. Murui et K. Wanaka, MEASUREMENT OF STERYLGLYCOSIDES BY HIGH-PERFORMANCE LIQUID-CHROMATOGRAPHY WITH 1-ANTHROYLNITRILE DERIVATIVES, Bioscience, biotechnology, and biochemistry, 57(4), 1993, pp. 614-617
A very sensitive and efficient high performance liquid chromtographic
(HPLC) method for measuring 0.5-50 ng of free and acylated sterylglyco
sides (FSG and ASG) in oils in the form of their 1-anthroylnitrile der
ivatives has been developed. Before to derivatization, the whole stery
lglycosides were simply isolated with silica gel cartridge then fracti
onated via thin layer chromatography (TLC) into pure FSG and ASG. Deri
vatization was done on the FSG molecular species, therefore ASG was co
nverted into FSG with mild alkaline hydrolysis. The derivatives separa
ted by reversed phase mode were detected at 254 nm. Avenasterylglycosi
de can not be separated from isofucosterylglycoside and campesterylgly
coside from stigmasterylglycoside. The method was successful with to d
ifferent vegetable oils commercially extracted with hexane as well as
those laboratory-extracted with a polar solvent mixture.
