A NEW APPROACH BASED ON THE LEAF DISC METHOD FOR AGROBACTERIUM-MEDIATED TRANSFORMATION AND REGENERATION OF ASPEN

A new protocol for increasing efficiency of production of transgenic a spen plants was developed. The new protocol was based on the leaf disc method but extended its power by inducing transformed roots with plan t growth regulator from leaf discs and then regenerating transgenic as pen plants from the transformed roots. This new protocol greatly impro ved recovery of transformed shoots, almost eliminated the occurrence o f escapes and might also reduce the occurrence of chimeric shoots. Thi s new protocol should be applicable to various plant species that can be infected by Agrobacterium and regenerated from roots. (C) 1997 Else vier Science Ireland Ltd.
