Micellar electrokinetic chromatography was used to resolve diastereome
rs of oligonucleotides possessing several chiral phosphoramidate bridg
es. These materials were not resolved by conventional liquid chromatog
raphic techniques. Parameters that were found to affect electrophoreti
c resolution included the nature and concentration of surfactant, pres
ence of organic modifiers such as propan-2-ol and urea, use of deuteri
um oxide-based buffers and temperature control. Practical optimization
was achieved by employing Terabe's resolution principles.
