The present study examines the possible existence of a mechanism regul
ating emptying of the secretory canaliculi content of the parietal cel
l and the possible effects of carbachol and gastrin on such a mechanis
m. In rabbit gastric glands stimulated with carbachol, C-14-aminopyrin
e accumulation reached a maximum after 15 min and then started to decr
ease. This decrease was not accompanied by a decrease in oxygen consum
ption, nor was any decrease of accumulated C-14-aminopyrine seen in di
spersed gastric cells. In glands but not in cells stimulated with hist
amine together with 3-isobutyl-1-methylxanthine (IMX), carbachol induc
ed a reduction in the accumulated C-14-aminopyrine content, whereas th
e effect of gastrin was less pronounced, The carbachol-induced reducti
on was counteracted by atropine but was not accompanied by a decrease
in oxygen consumption. It is suggested that there exists a mechanism t
hat controls the emptying of the secretory canaliculi content of the p
arietal cell, and that carbachol, in addition to stimulating acid prod
uction, also contributes to this emptying. Paracrine factors may be in
volved in this latter mechanism.
