The combination of high-resolution scanning, image-databasing technolo
gy and sequence assembly software makes it possible to assemble contig
uous overlapping sequences of DNA in a fraction of the tune required b
y manual methods. This paper describes and evaluates an assembly progr
am that rapidly generates contigs from scanned images and provides a u
nique ability to verify disagreements between overlapping strands or f
ragments of complementary DNA sequences on-screen, further increasing
sequencing throughput.
