Buffalo genomic DNA digested with Pst I restriction endonuclease revea
led 5 conspicuous bands of repetitive DNA fragments by horizontal agar
ose-gel electrophoresis. The molecular sizes of the fragments were 0.5
,0.7, 1.25, 1.35 and 2.1 kbs. Buffalo genomic DNA - Pst I digests hybr
idized with cattle -Eco RI 1.4 kb repetitive DNA fragment even under h
igh stringent condition, hence, revealed high degree of sequence homol
ogy between satellites of buffalo and cattle. Southern hybridization p
atter revealed bands of 0.69, 1.39, 2.11, 2.75 and 3.43 kbs and thus d
emonstrated a periodicity of approximately 700 bp, similar to that obs
erved in cattle. Agarose-gel electrophoresis and southern blot hybridi
zation revealed sequence homology and similar periodicity of 700 bp be
tween satellites of buffalo and cattle.
