The introduction of the confocal laser scanning microscope makes it po
ssible to acquire fluorescent specimens in 3D. We present basic image
processing tools to enhance the data and to quantitate the morphology
and topography of the intravolume elements. The tools were applied to
the description of the spatial distribution of DNA replication sites i
n mammalian cell nuclei as an example.
