H-Phosphonate synthesis of oligodeoxynucleotides is described in which
acetonitrile is replaced by dichloromethane. This solvent is thereaft
er easily regenerated and reused. Practically ''one solvent'' procedur
e of oligonucleotide synthesis has been established. The preparation o
f -N2-(N,N-dimethylaminomethylene)-2'-deoxyguanosine (Ic) and -N6-(N,N
-dimethylaminomethylene)-2'-deoxyadenosine (Id) is described.
