Fatty acid methyl esters suitable for gas chromatographic analysis wer
e prepared by treatment of bacterial cells with sodium methoxide (1 N)
in anhydrous methanol. The methanolysis of cells, washed or not with
sterile water, was carried out at room temperature for 5 min. This met
hod was effective for Gram-positive and Gram-negative bacteria, but no
t for growing yeast cells. The coefficient of variation of fatty acid
methyl ester values was 4.7% of the mean values (range 0.4-25%).
