The siderophores produced by Pseudomonas fluorescens and P. chlororaph
is were detected from the culture supernatants in MM9 and modified Kin
g's medium by the universal CAS assay at wavelengths 620-690 nm. The C
AS assay was applied to detect Pseudomonas siderophores directly in si
tu, during their production phase, in modified King's medium. Optimum
results were detected with a final CAS concentration of 0.025 mM and a
n iron concentration of 1.25 muM. The problems of the method are discu
ssed with respect to the absorbance spectrum, the toxicity of the HDTM
A detergent, the influence of the iron concentration and the complexit
y of media for siderophore production.
