Acw. Ward et al., FLOW-CYTOMETRIC ASSESSMENTS OF PLOIDY STABILITY IN CRYOPRESERVED DIHAPLOID SOLANUM-TUBEROSUM AND WILD SOLANUM SPECIES, Cryo-letters, 14(3), 1993, pp. 145-152
Ploidy stability in dihaploid cultivars of Solanum tuberosum and in wi
ld genotypes, regenerated from cryopreserved shoot-tips, was assessed
by flow cytometry. Frozen shoot-tips were compared with explants of po
tato subjected to other in vitro manipulations. The frequency of polyp
loidization was minimal in plants regenerated from cryopreservation as
compared with those derived from leaf explants and protoplasts. Cryop
reservation does not appear to induce ploidy changes in sensitive diha
ploids of S. tuberosum.
