Changes in ATP contents in cultured rat cerebellar granule cells and m
ouse hippocampal neurons during and after glutamate (Glu) treatment we
re measured using the luciferase-luciferin method. After a 15-min Glu
treatment (50 muM, 0 Mg2+), the ATP contents (in nmol/ng of protein) d
ecreased approximately twofold, and remained at this or even lower lev
el after a 430-min washing of cultures by control solution. This resul
t is of importance for understanding the origin of Glu-induced destabi
lization of Ca2+ homeostasis, since both Ca2+-extruding systems, Na(o)
+/Ca(i)2+ exchange and Ca2+ pump, are known to be ATP - dependent.
