The in situ techniques for DNA and RNA colony hybridization were adapt
ed for application to Escherichia coli bacterial cells grown in liquid
media and loaded onto membrane filters by microfiltration. A linear c
orrelation was found between the amount of loaded cells and the P-32-r
adioactivity retained on the filters. This makes it possible to apply
both techniques for quantitative determination of specific polynucleot
ides produced at different stages of bacterial growth.
