Key enzymes of the glyoxylate cycle, isocitrate lyase (ICL) and malate synt
hase (MS), have been detected in the liver of alloxan-treated rats. The act
ivity of ICL in rat liver was 0.040 mu mol/min/mg protein and the activity
of MS was 0.022 mu mol/min/mg protein. These enzymes were associated with t
he peroxisomal fraction, The activities of citrate synthase, malate synthas
e and malate dehydrogenase detected in the peroxisomal fraction were also i
ncreased by alloxan treatment. Isocitrate lyase was partially purified and
displayed catalytic and regulatory properties similar to those of the enzym
e isolated from the liver of starved rats (Popov, V.N. et al. (1996) FEBS L
ett. 391, 87-90). (C) 1998 Federation of European Biochemical Societies.
