Kinin B-1 receptors are induced by various inflammatory mediators. The aim
of the present study was to investigate the effect of the CXC chemokine IL-
8 on kinin B-1 receptor expression in IMR-90 cells, by performing binding s
tudies and Northern blot analysis of B-1 receptor mRNA levels. We demonstra
ted here that the density of the kinin B-1 receptors could be increased by
the chemokine IL-8 in a concentration- and time-dependent manner. IL-8 also
increased the kinin B-1 receptor mRNA level in IMR-90 cells. IL-8-induced
B-1 receptor expression could be totally abolished by pretreatment with the
metabolic inhibitors. Furthermore, expression was markedly reduced by anti
bodies to human IL-1 beta. In conclusion, IL-8 increased the expression of
kinin B-1 receptors in IMR-90 cells and this effect is likely to be seconda
ry to the production of IL-1 beta. (C) 1998 Academic Press.
