New cloning vectors with temperature-sensitive replication

A series of cloning vectors with conditional, temperature-sensitive replica tion that are selectable with ampicillin, chloramphenicol, and kanamycin ha s been constructed. These vectors are derivatives of a pSC101 mutant that c an replicate only at low temperatures. The cloning vectors carry a number o f unique restriction sites and provide for screening of recombinant plasmid s by ct complementation. These vectors have proven useful for a variety of applications where conditional replication of a recombinant plasmid is desi red. (C) 1999 Academic Press.
