A rapid and sensitive method was developed for the analysis of disaccharide
composition in heparin (HP) and heparan sulfate (HS) by reversed-phase ion
-pair chromatography on a 2 mu m porous silica gel column. HP and HS were d
igested with heparin lyase I, II and III in combination, and the produced u
nsaturated disaccharides were separated within 15 min. Calibration graphs w
ere linear in the range 1 ng-1 mu g with the fluorometoric post-column dete
