Time-dependent pH-change in halobacteria was observed with fluorescent
dye. Mechanical improvement of a cell holder in a fluorescence spectr
ometer enabled us to measure the intracellular pH-change that proved d
ifferent from the extracellular change. Upon irradiation, the extracel
lular pH of the strain initially changed into higher value and subsequ
ently reverted to lower value until irradiation ceased, whereas the in
tracellular pH monotonously increased for freshly prepared cells. Neve
rtheless, the pH-changes in liposome with bacteriorhodopsin were compl
ementary each other.
