Dextrin prepared from corn starch was immobilized on kieselguhr or chi
tin using a crosslinking reagent (epichlorohydrin), to produce a new a
dsorbent having a high affinity for alpha-amylase. The dextrins were i
mmobilized at levels of 300-340 and 200-270 mg/g adsorbent on chitin a
nd kieselguhr, respectively. There was an optimum crosslinking time fo
r alpha-amylase adsorption because of a balance between the number of
adsorption sites and the affinity of the immobilized dextrin for alpha
-amylase. Adsorption of alpha-amylase on the crosslinked immobilized d
extrin was as much as 50 times higher than the value on raw corn starc
h. When expressed in terms of the content the adsorption of alpha-amyl
ase varied with the type of support (kieselguhr and chitin) because of
the different state of the immobilized dextrin. (C) 1997 Published by
