A series of hydroxylated benzylideneanilines and benzylamines were pre
pared and tested for inhibition of epidermal growth factor receptor-as
sociated protein tyrosine kinase (PTK) activity in vitro. Molecular mo
delling and analysis of the biological results lead us to propose a mi
nimal structural pharmacophore for two distinct binding sites within t
he PTK domain.
