Abstract
Substitution of Glu for the evolutionarily ! conserved Asp-97 in the a ctive site of Escherichia coil inorganic pyrophosphatase increases the apparent pK(a) of the essential acidic group controlling the catalyti c constant for pyrophosphate hydrolysis!. In combination with previous ly reported data, this fact suggests that activity!. decreases in alka line medium because of decreased rats of the rt release of the first m olecule of the product phosphate from the active site.
