A simple method is described for detecting glycoproteins which had bee
n dot-blotted or electro-blotted on to a polyvinylidene difluoride (PV
DF) membrane after sodium dodecyl sulfate (SDS)-polyacrylamide gel ele
ctrophoresis. This method is based on the periodate oxidation of glyco
proteins on a PVDF membrane with subsequent staining by the chromophor
ic hydrazide, 4-N,N-dimethylamino-4'-azobenzene sulfonyl hydrazide. Th
e glycoproteins could be visualized as red-colored spots or bands in t
he range of 0.16-0.31 mu g.
