The effect of alpha-lipoate and dihydrolipoate on the mitochondrial pe
rmeability transition was investigated. Both substances promoted the p
ermeability transition in isolated rat liver mitochondria and in perme
abilized hepatocytes, dihydrolipoate most potently in spite of it bein
g a dithiol. The stimulation was prevented by Cyclosporin A or hydroxy
butyltoluene but not by ascorbate. It is suggested that the greater po
tency of dihydrolipoate could be due to formation of a radical.
