Dj. Gillooly et Ca. Fewson, PRODUCTION OF A PERILLYL ALCOHOL-DEHYDROGENASE BY SITE-DIRECTED MUTAGENESIS OF A BENZYL ALCOHOL-DEHYDROGENASE, Biotechnology letters, 20(3), 1998, pp. 325-327
Benzyl alcohol dehydrogenase from Acinetobacter calcoaceticus oxidises
a wide range of aromatic and other cyclic alcohols and it has high sp
ecificity constants for these substrates, but it does not oxidise shor
t- or long-chain aliphatic alcohols. Mutation of an active-site argini
ne to a histidine can switch the substrate specificity of the enzyme s
o that it has a very much greater preference for perillyl alcohol than
for benzyl alcohol.
