J. Zbaeren et al., A NEW METHOD FOR GMA SECTIONS - IMMUNOFLUORESCENCE WITH THE ELF-PRECIPITATE COMBINED WITH A CLASSICAL HEMATOXYLIN EOSIN-PHLOXIN STAIN/, Journal of histotechnology, 21(1), 1998, pp. 25-27
Rat pancreas, embedded in 2-hydroxyethyl methacrylate (GMA), was used
to demonstrate glucagon by immunofluorescence in combination with a he
matoxylin/eosin-phloxin (H/E-P) stain. This method for GMA embedded ti
ssue enables a good differentiation with both light and fluorescence m
icroscopy. The enzyme-labeled-fluorescence (ELF) precipitate marked im
munoreaction can be visualized with UV excitation, whereas the H/E-P s
tain can be observed in both bright field and with blue or green excit
ation. The ELF-signal together with the surrounding tissue structure c
an be visualized with double bandpass filters that enable UV together
with blue or green excitation. Alternatively a similar effect can be a
chieved photographically by double exposure.
