We prepared a monoclonal antibody (mAb), IgM class, to rabbit muscle a
ldolase (ALD) A(4). The mAb reacted with the ALD A subunit but not wit
h the B and C subunits. Distribution of the ALD A subunit was studied
employing immunohistochemistry. with the ABC procedure in the rabbit t
issues. The ALD A subunit was localized in the I-band of the skeletal
muscle fibers. In the liver, endothelial cells showed strong immunorea
ctivity while parenchymal cells were negative or yielded only a faint
reaction. The collecting tubules were positively stained in the kidney
. Strong reactivity was seen in the proximal tubules of the outer medu
lla but not in those of the cortex. In the adrenal gland, cells in the
cortex exhibited strong immunostaining, while medullary cells were ne
gative. In the testis, most of the seminiferous epithelium was moderat
ely immunoreactive. Intense staining was present in interstitial cells
. The ALD A subunit was localized in the interstitial cells and follic
ular cells of primary follicles in the ovary. In the cerebellum, the A
LD A subunit was localized exclusively in nerve fibers of the white ma
tter; Purkinje cells and nerve cells in the granular layer were negati
ve.
