The effects of TDZ (N-phenyl-N'-1,2,3-thidiazol-5-ylurea) and CPPU (N-
(2-chloro-4-pyridil)-N'-phenylurea) on bud initiation of Amaranthus ed
ulis thin cell layers (tTCLs) explants were investigated. tTCLs (0.2-0
.4 mm thick) excised from 1 week old seedlings were cultured on Murash
ige and Skoog (MS) basal salts medium supplemented with varying concen
trations of TDZ or CPPU ranging from 1 to 20 mu M and 1 week after cul
ture, buds were initiated. The optimal concentrations were 2 mu M for
TDZ and 10 mu M for CPPU. Buds developed into normal plants after tran
sfer onto a MS basal salts medium containing BA to obtain normal inter
node elongation and onto a MS basal salts medium containing IBA or TDZ
for rooting. (C) 1998 Elsevier Science Ireland Ltd. All rights reserv
