Abstract
Prostaglandin E-1 has been clinically used in a variety of vascular oc clusive diseases. We investigated the in vitro effect of prostaglandin E-1 on the synthesis of tumor necrosis factor-alpha, interleukin-1, i nterleukin-6, interleukin-10 and transforming growth factor beta by hu man peripheral blood mononuclear cells stimulated with lipopolysacchar ides. Prostaglandin E-1 significantly suppressed both the mRNA express ion and the production of tumor necrosis factor-alpha and interleukin- 10 by lipopolysaccharides-stimulated peripheral blood mononuclear cell s in a dose-dependent manner (1X10(-6)-1x10(-8) M). Since tumor necros is factor-alpha is a potent proinflammatory cytokine involved in certa in inflammatory diseases, our findings suggest the possibility to expa nd the clinical application of prostaglandin E-1.
