A new process for the purification of protein A from Staphylococcus au
reus has been developed achieving purity of more than 98% with a recov
ery of 30%. The process involves extraction employing a specific buffe
r containing Toluene-Triton X-100-EDTA and subsequent purification usi
ng macroporous glycidyl copolymer. The process has been successfully u
sed to purify protein A of both extracellular and intracellular origin
. The process is simple, rapid, inexpensive and efficient when compare
d to many other reported processes. (C) 1998 Elsevier Science Ltd.
