A. Wise et al., INTERACTION OF THE G-PROTEIN G(11)ALPHA WITH RECEPTORS AND PHOSPHOINOSITIDASE-C - THE CONTRIBUTION OF G-PROTEIN PALMITOYLATION AND MEMBRANEASSOCIATION, FEBS letters, 407(3), 1997, pp. 257-260
Wild-type and palmitoylation defective mutants of the murine G protein
G(11)alpha were transfected into HEK293 tells. Wild-type G(11)alpha w
as membrane associated, Cys9Ser Cys10Ser G(11)alpha was present in the
soluble fraction whilst both Cys9Ser G(11)alpha and Cys10Ser G(11)alp
ha were distributed between the fractions. Expression of the rat TRH r
eceptor resulted in agonist stimulation of inositol phosphate accumula
tion. The degree of stimulation produced by TRH following co-transfect
ion of the palmitoylation-resistant forms of G(11)alpha compared to th
e wildtype protein correlated with the amount of membrane-associated G
protein. (C) 1997 Federation of European Biochemical Societies.
