Abstract
The wheat monosomics 1A, 1B, 1D and monotelodisomics 1AL, 1BL, 1DL ind ividuals were identified by electrophoretic analyses of high molecular weight glutenin subunits (HMW-GS) and gliadin blocks combined with qu antitative densitometric evaluations of glutenin and gliadin profiles. Absence of a whole chromosome or chromosome arm was confirmed by comp arison of relative quantity of corresponding HMW-GS or gliadin block e xpressed in aneuploid and euploid grains. Decreased quantity of expres sed protein is detectable in electrophoretic gels and is connected wit h absence of the corresponding chromosome or chromosome arm, i.e., wit h absence of the locus (or loci) controlling production of HMW-GS or g liadin proteins. The HMW-GS are useful molecular markers for simple, f ast, and unambiguous identification of wheat monosomics 1A, 1B, 1D, or monotelodisomics 1AS, 1BS, 1DS. Quantitative analysis of gliadin prof iles can facilitate identification of absent shea chromosome arms in m onotelodisomics 1AL, 1BL, and 1DL. Gliadin analyses can be used for id entification of monosomics 1A of wheat genotypes possessing the null a llele at locus Glu-A1.
