Insights into tolerance and autoimmune processes have led to novel immunoth
erapeutics for inhibiting autoimmune disease in animal models. However, rec
ent studies question the immune basis of some of these therapeutic strategi
es and raise concerns about their efficacy and safety. Here, we discuss the
feasibility of extending the success of antigen-based immunotherapeutics f
or T-cell mediated autoimmune diseases from animal models to humans.
