Pharmacokinetic studies of cefazolin (CFZ) were conducted after single dose
(30 mg/kg, IV) in afebrile goats and febrile goats with and without proben
ecid (1 g, IV). The zero time plasma concentration (C-p(0)) in febrile goat
s with probenecid(PB) was higher (p < 0.05) compared with the febrile goats
-without PB. The PB administration prolonged the time of the therapeutic pl
asma concentration (Cp ther) in the febrile group with PB as compared to wi
thout PB. Concurrent administration of PB with CFZ decreased (p < 0.05) the
values of total body clearance (cl(B)), renal clearance (cl(R)) and hepati
c clearance (cl(H)) Of CFZ in febrile goats. Volume of distribution is usua
lly expected to change when a febrile state is induced. Lack of such an eff
ect in this study could be due to high individual variation. PB could block
tubular secretion of CFZ and prolong the Cp ther in febrile goats when giv
en concurrently with CFZ. It is concluded that probenecid may be given IV w
ith CFZ especially when higher plasma concentrations of CFZ are required fo
