Seven nucleotides linked through the gamma-phosphate to diamine hydroc
arbons were synthesized and coupled to Sepharose for use in protein pu
rification affinity chromatography. The synthesis involved converting
the nucleotides to nucleoside-5'-trimetaphosphates using dicyclohexyl
carbodiimide, followed by nucleophilic ring opening of the trimetaphos
phate with an alpha omega-diamino hydrocarbon to generate a gamma-phos
phoamide linkage in each nucleotide.
