Lily (Lilium x formolongi hort.) protoplasts were isolated from merist
ematic nodular cell clumps using enzyme solution containing different
concentrations of silver thiosulfate (STS), embedded in 0.1% (W/V) gel
lan gum - solidified medium containing half strength mineral salts of
Murashige and Skoog (1962) medium, 4.1 mu M 4-amino-3,5,6-trichloro-pi
colinic acid (picloram) and 0.5 M glucose, and cultured at 25 degrees
C in the dark. The highest plating efficiency (17.7%) was obtained whe
n the protoplasts were isolated using the enzyme solution containing 1
0 mu M STS.
