Truncation of human chromosomes at desired sites by homologous recombi
nation techniques enables functional and structural analyses of human
chromosomes and development of human artificial chromosomes. However,
this targeted truncation has been inefficient. We describe here an eff
icient method for targeted truncation in the chicken DT40 cells with a
high homologous recombination rate. The human chromosome 22 was trans
ferred into DT40 cells, where human telomeric repeat (TTAGGG)n was tar
geted to the LIF locus on the chromosome. Molecular and cytogenetic an
alyses showed that the predicted truncation at the LIF locus occurred
in all of the targeted clones.
