A conjugate of poly(gamma-glutamic acid)(gamma-PGA) with 5-fluorouraci
l (5-FU) ( ly(alpha-hydroxymethyl-5-fluorouracil-glutamate)), gamma-PF
UG, was prepared as a polymeric prodrug matrix. The release of 5-FU an
d the degradation behavior of gamma-PFUG, which is soluble in polar or
ganic solvents were studied. gamma-PFUG films were prepared by casting
from DMF. The gamma-PFUG matrix degraded in 7 days and incorporated 5
-FU was released in 3 days. 5-FU release was accelerated by mixing fre
e 5-FU into gamma-PFUG. Here, gamma-PFUG functioned as a hydrophobic d
egradable matrix. Analogous with gamma-PFUG, gamma-PGA conjugated with
a drug may be useful as a biodegradable polymeric prodrug.
