The enzyme polyamine oxidase from maize has been purified and covalent
ly immobilized onto polymeric supports to assemble a polyamine electro
chemical biosensor. The enzyme has been used in conjunction with eithe
r an O-2 or a H2O2 electrode. The analytical behaviour of this enzyme
electrode has been studied with respect to sensitivity, selectivity, o
ptimum pH and buffer, response time and lifetime. Recovery studies per
formed on urine samples demonstrated that the polyamine biosensor deve
loped can be successfully used to measure polyamines in biological flu
ids.
