A procedure, adaptable to large-scale remote operation, was developed
to purify no-carrier-added (NCA) (SC)-S-47 from irradiated Ti targets.
Methods based on extraction chromatography and cation-exchange chroma
tography were compared. Results of this comparison led to the developm
ent of an optimized procedure based on cation-exchange with Dowex AG 5
0W-X4 and Sc-47 elution with HCl/HF. This method gave 90-97% overall S
c-47 recovery, with a Ti separation factor greater than 2.4 x 10(-5),
and specific activities greater than or equal to 0.9 GBq mu g(-1). Use
of the Sc-47 product, for labeling monoclonal antibodies, resulted in
consistent labeling yields of greater than or equal to 90%. (C) 1998
