The chemical approaches to the study of the DNA template biosynthesis
by DNA polymerases and reversed transcriptases are considered with the
main emphasis on E. coli DNA polymerase I and HIV-1 reversed transcri
ptase. Special attention is paid to the comparison of the chemical mod
ification, X-ray analysis data, as well as to the quantitative aspects
of the DNA polymerases interactions with templates, primers and nucle
oside-5'-phosphates. New versions of the catalytically competent label
ling developed in the course of investigation of these enzymes are des
cribed. The results of application of the primer analogues with photor
eactive groups to the study of the replication complexes are presented
