An enzyme-linked immunosorbent assay (ELISA), based on polyclonal anti
bodies, was used to quantify didecyldimethylammonium chloride (DDAC) i
n industrial antisapstain treating solutions. The initial ELISA valida
tion included: cross-reactivity with similar analytes, interference wi
th non-related compounds, percentage of false negative and false posit
ive determinations, and comparison of the immunoassay data with the da
ta from titration and HPLC methods. ELISA and the titration method gav
e comparable results. However, higher DDAC values were obtained for in
dustrial treating solutions measured by ELISA than by HPLC. Immunoassa
ys may be a potential analytical tool for the quality control program
of DDAC type antisapstain formulations, if specificity could be improv
ed.
