Abstract
A method based on differences in electrophoretic mobility of RNA trans cripts made from polymerase chain reaction (PCR) products was used for differentiation among virus isolates. A T7 RNA polymerase promoter wa s attached to amplified prunus necrotic ringspot virus (PNRSV) sequenc es by PCR. The PCR products then served as a template for transcriptio n. Single-stranded transcripts originated from different PNRSV isolate s varied in electrophoretic mobility in polyacrylamide gels, presumabl y because of transcript conformation polymorphism (TCP).
