Mutation and damage in DNA by interaction, UV-irralation, nitrite acti
on and ultrasonication were studied by means of voltammetry using a gr
aphite electrode covalently immobilied with ssDNA-1, Daunomycin was us
ed as a hybidization indicator. When the complementary ssDNA-2 in solu
tion was damaged by phsical or chemical agents, the hybridization of s
sDNA-1 and ssDNA-2 was changed, All the results in voltammogram indica
ted that intercalator can't destroy the double-helical structure of DN
A, while UV-irralation and nitrite action can make the permanent damag
e, Ultrasonication can reduce polymerization in DNA and the hybridizat
ion time was shortened. This may provide a new method for clinical med
icine study.
