S. Popiel et al., THIN-LAYER CHROMATOGRAPHY AND ENZYMATIC ANALYSIS OF PHOSPHORORGANIC COMPOUNDS USING 4-METHYLUMBELLIFERONE ESTERS, Chemia Analityczna, 43(4), 1998, pp. 733-742
The procedure of phosphororganic compounds analysis using thin layer c
hromatography with enzymatic method of chromatograms visualization was
elaborated. As the substrates of enzymatic reactions, using cholinest
erase, esters of 4-methylumbelliferone were applied. The effects of re
actions obtained when 4-methylumbelliferone esters were used were comp
ared with the effects of reactions when indoxyl acetate was used. The
performance of enzymatic reaction on a chromatographic plate was estim
ated by the fluorescence measurement of the reaction product, which wa
s 4-methylumbeliferone. The enzymatic reaction was performed by pressi
ng the papers saturated with proper reagent to the chromatographic pla
te. It was the way to eliminate from enzymatic reaction medium organic
solvents used for dissolving 4-methylumbeliferone eaters. Using the p
rocedure it was possible to determine sarin, soman and tabun at the le
vel of dozen picograms per spot and VX as well as pesticides from the
individual nanograms to their dozens per spot.
