Two high-performance Liquid chromatographic (HPLC) methods are describ
ed for determination of (+/-)-ethopropazine (ET) in rat plasma. After
deproteination and Liquid-Liquid extraction, assay of (+/-)-ET was per
formed using either a C-18 column (non-stereospecific assay) or an (al
pha-R-naphthyl)ethylurea column (stereospecific assay). The UV detecti
on was at 250 nm. Mean recovery was >85%. Both assays demonstrated exc
ellent linear relationships between peak height ratios and plasma conc
entrations; quantitation limits were less than or equal to 25 ng/ml, b
ased on 100 mu l rat plasma. Accuracy and precision were <17% with bot
h methods. Both methods were applied successfully to the measurement o
f ET plasma concentrations in rats given the drug intravenously. (C) 1
