A full-factorial design was used to assess the influence of ion streng
th and Triton X-114 concentration on the extractive purification of a
lipase from Acinetobacter calcoaceticus AAC 323-I in temperature-induc
ed aqueous two-phase systems. The condition for optimal extraction was
found at 0.75 M NaCl and 3% Triton X-114 concentrations, as shown by
surface response analysis. Based on these results, a rapid lipase puri
fication procedure from culture supernatant was developed. Such proced
ure allows direct extraction from culture broths in three successive s
teps with no additional operations. This technique enabled us to extra
ct 68% of the enzyme and achieved a purification factor of 41.
