An immunoelectrochemical assay in combination with homogeneous enzyme-
labeled antibody conjugation was developed for rapid detection of Salm
onella. The assay was performed by mixing alkaline phosphatase linked
anti-Salmonella (APLAS) with Salmonella in a solution. The Salmonella-
APLAS conjugate separated by polycarbonate membrane filtration was inc
ubated with a phenyl phosphate substrate to produce phenol. The concen
tration of Salmonella cells was determined by measuring phenol oxidati
on peak current using differential pulse voltammetry at a renewable ca
rbon paste electrode. This assay could be completed within two hours w
ith a detection limit of 5 x 10(3) cells/mL. A linear response was fou
nd for Salmonella between 5 x 10(3) and 1 x 10(6) cells/mL.
