Protein kinase G (PKC), which has a wide variety of functions, compris
es at least twelve isoenzymes that have distinct roles respectively. R
esearch in recent years indicates that PKC zeta plays a pivotal role i
n the regulation of cell proliferation. In order to investigate its ro
le in the regulation of keratinocyte proliferation, eukaryotic express
ion plasmid of anti-sense PKC zeta is constructed and transfected into
human keratinocyte line, Colo16. The human keratinocyte model express
ing anti-sense PKC zeta is established successfully, the growth proper
ties of the cell model is analyzed by the detection of cell growth cur
ve and cell cycle phase. The mechanism of the relationship between PKC
zeta and CyclinD1 is also investigated by the western blotting analys
is of CyclinD1 protein.
